Very interesting approach on using immune checkpoint inhibition as a treatment for progressive multifocal leukoencephalopathy (PML). Keytruda (pembrolizumab) demonstrated efficacy in improving or stabilizing PML, lowering JC viral load, and enhancing anti-JC virus CD4 and CD8 T-cell activity.

This is good news for patients who receive potent immunosuppresive or immunomodulatory therapies (eg, Tysabri for multiple sclerosis) and are at risk for PML.

It would be interesting to look at responders vs nonresponders and determine if there are specific patient or disease factors that correlate with Keytruda responsiveness.

These immune checkpoint inhibitors are now revolutionizing the treatment of cancer and saving millions of lives.

I met Dr Allison years ago and could not think of someone more deserving.
